Epitalon is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) developed in Russia as a synthetic analog of epithalamin, a bovine pineal-gland extract. It is studied in longevity research for reported telomerase activation, telomere maintenance, and circadian/melatonin regulation. Nearly all published research originates from a single Russian research group, and it has not undergone Western-standard randomized controlled trials. It is not FDA-approved.
Epitalon (also spelled Epithalon) was developed by Vladimir Khavinson and colleagues at the St. Petersburg Institute of Bioregulation and Gerontology, based on the active components of epithalamin. It is a short tetrapeptide with sequence Ala-Glu-Asp-Gly (AEDG) and molecular weight ~390.4. Its most-cited mechanism is activation of telomerase (induction of hTERT expression) with reported telomere elongation in human somatic cell cultures, alongside effects on melatonin rhythm and antioxidant systems. Khavinson-group studies in animals and elderly human cohorts have reported geroprotective effects and reduced mortality, but independent Western replication is limited, and the single-group dominance of the literature is a major caveat. Mechanism of Action
Reported to activate telomerase by inducing expression of hTERT (the catalytic subunit of telomerase), supporting telomere length maintenance; also reported to normalize circadian/melatonin rhythms and exert antioxidant and neuroendocrine effects. Mechanistic data come largely from in vitro and animal studies.
